Microbiosci announced the release of its Bacillus Subtilis Genome Editing Service to support gene knockout, gene insertion and point mutation for either research or industrial purposes.

New York, USA – May 27, 2021 – Microbiosci, the mature division of Creative Biogene, is fully engaged in developing unique technologies to facilitate the research in the field of pharmaceutical, biotechnology, agricultural, and diagnostics industries. With years of experience, Microbiosci develops a wide range of high-quality products and services in microbial genome editing, identification, sequencing, proteomics and metabolomics. Recently, Microbiosci announced the release of its Bacillus Subtilis Genome Editing Service to support gene knockout, gene insertion and point mutation for either research or industrial purposes.

Bacillus subtilis is a kind of Gram-positive bacteria with good characteristics and has been widely used in the production of heterologous proteins. Bacillus subtilis and some of its close relatives have excellent protein secretion capabilities and are generally considered safe (GRAS), making them an important host for the production of antibiotics, medicinal proteins and industrial enzymes. Microbiosci’s Bacillus subtilis genome editing service is based on CRISPR/Cas9 technology and homologous recombination technology:

Red / ET recombination can use phage protein pair (RecE / RecT or Reda / Redb) mediated homologous recombination to carry out DNA engineering in Bacillus subtilis. The core step of Red / ET recombination is the crossover step between the targeting construct containing homology arms and the target. The target can be the locus of Bacillus subtilis. By designing a homologous fusion fragment of the target gene, clone it to suicide. In the vector, the suicide vector is then transformed into the target bacteria. The insertion mutants were selected by antibiotic screening. Under the second round of negative selection pressure, only mutations that include the second homologous recombination and suicide plasmid loss can be spared. Through PCR screening and sequencing, researchers can obtain mutants of this strain. The specific workflow includes homology arm design and suicide plasmid construction, recombination, selection/screening, verification, etc.

CRISPR technology is derived from the immune system in bacteria and archaea. It is an effective genome-scale editing tool. It has completely changed the traditional genetic engineering methods and promoted strain engineering as never before. It enables Bacillus subtilis to perform genetic manipulations quickly and reliably. Two components are needed to work: a guide RNA (gRNA), such as under the RNA polymerase III promoter, and a DNA endonuclease fused with a nuclear localization tag, of which Cas9 is the most commonly used. The project process includes sgRNA design and construction, transformation, selection/screening, and verification.

“The Bacillus subtilis genome editing service provided by Microbiosci mainly helps gene destruction, deletion or insertion; reporter gene and tag integration; promotion of fine-tuning; introduction of point mutations,” said Marcia Brady, the marketing director of Creative Biogene, she also said, “Creative Biogene can provide the best services for Bacillus subtilis genome editing. Our talented experts are dedicated to editing your Bacillus subtilis genome for the greatest chance of success.”
